Mole:

12 strangers work together to complete various tasks each worth varying amounts of money that go into a pot. Sometimes you can even lose money if you fail a task or part of a task. Among this group of strangers is the Mole, hired by the show to sabotage the group's efforts. The others must figure out who the Mole is and learn as much about them as possible (what they did in each task, sometimes what they wore, where they sat at dinner etc.). Each round there is a quiz on the identity of the Mole. Whoever scores lowest (ie knows the least about the Mole) is eliminated. Along the way various temptations to screw over the others are often offered, mostly in the form of exemptions from the next quiz (ie Immunity) if they agree to screw something up thus costing the group money. At the end there are 3 players (2 real contestants and the Mole). Whoever answers the most correctly in the final (longer) quiz wins. In cases of ties it's whoever answered the quiz fastest. The winner wins however much is in the pot by the finale.

A common strategy amoung real players is to look like you're the Mole by sabotaging tasks either overtly or somewhat covertly and make people answer their quizzes for you being the Mole, thus increasing your odds of answering more questions correctly. Players often form alliances (or coalitions) to share information on who they think is the Mole and divert other players away from their suspect. It's a game of observation, reading people and intuition.

There are usually hidden clues in the show for viewers at home to try and figure out.
